The Role
Accurate pharmacokinetic modeling is essential in preclinical drug development, particularly for biologics like therapeutic antibodies. Physiologically based pharmacokinetic (PBPK) models play a pivotal role in predicting Target-Mediated Drug Disposition (TMDD), guiding optimal dosing strategies and ensuring therapeutic efficacy and safety. At Genmab, we leverage quantitative approaches, including PBPK modeling, to better understand the pharmacokinetics of antibody-based therapeutics and their interactions with different targets to advance drug candidates.
As an intern within the Quantitative Translational Pharmacology team at Genmab, you will contribute to PBPK modeling projects. Your responsibilities will include analyzing and integrating diverse target expression datasets, including multi-level datasets such as protein abundance, mRNA expression levels, and immunohistochemistry (IHC) scoring and quantitative immunofluorescence, to support robust PBPK model development. Understanding the localization of therapeutic targets across different tissues and organs will be a crucial component of your analysis. You will gain practical experience in data handling, quantitative modeling, and scientific communication within a dynamic biotech environment. Responsibilities
- Collecting comprehensive target expression data from public databases, proprietary resources, and multi-level datasets (protein abundance, mRNA expression, IHC scoring, quantitative immunofluorescence).
- Developing computational tools, leveraging methods such as the sigmoidal conversion approach by Sepp & Muliaditan (2024), to integrate heterogeneous datasets into accurate and standardized target concentration estimates.
- Incorporating target localization information to enhance the accuracy and relevance of PBPK model predictions.
- Validating PBPK model predictions by comparing simulated outcomes against experimentally observed preclinical pharmacokinetic data for known therapeutic targets (e.g., EGFR, HER2).
- Effectively communicate results and insights through concise reports and clear presentations.
This internship provides an opportunity to engage directly with applied quantitative modeling in drug development, offering flexibility to tailor specific project directions according to your interests and expertise!
Requirements
- Enrolled in a Master’s program in Statistics, Biopharmaceutical Sciences, Biomedical Sciences, Drug Innovation, Pharmacy, Data Science or a related field at a University in the Netherlands.
- Experience with R programming.
- Basic knowledge of cancer biology and/or immunology.
- You have experience in, or a strong interest in learning, single-cell data analysis and antibody drug discovery.
- You enjoy working collaboratively in a multidisciplinary team.
- Excellent communication and collaboration skills in English.
- Previous programming experience, ideally with the development of your own package.
Preferred Qualifications, Capabilities, and Skills
- Knowledge of AI techniques applied to chemical compound characterization, target discovery, and single-cell data analysis.
- Experience with biomedical or chemical research datasets.
- Familiarity with immuno-oncology, gynecological cancers.
- Previous industry experience or internship in a related field.
General Internship Information
- Internship duration: 6-9 months.
- Start date: September 1st / October 1st.
- Location: Utrecht, Netherlands
- Hybrid schedule: at least 3 days in the office, 2 days remote per week.
